Try to load it in safe mode directly and check windows event log.
Also try adding. Net 3.x from the additional feature.

Funny thing. After doing a fresh install of 15063, DDU worked once. But now, it won't run at all. But I'll confirm the above. It does work if you reboot into safe mode.

I checked the Event viewer logs and got two error messages.

    First -

    Application: Display Driver Uninstaller.exe
    Framework Version: v4.0.30319
Description: The process was terminated due to an unhandled exception.
Exception Info: exception code c0000005, exception address 00007FFC2BAAFD0C

    And Second -

Faulting application name: Display Driver Uninstaller.exe, version: 17.0.6.1, time stamp: 0x58cbe74c
Faulting module name: d3d9.dll, version: 10.0.15063.0, time stamp: 0x631de416
    Exception code: 0xc0000005
    Fault offset: 0x000000000000fd0c
    Faulting process id: 0x2170
    Faulting application start time: 0x01d2a986bb0e3a0d
    Faulting application path: E:\+Apps And Patches+\+Patches+\+Drivers+\Display Driver Uninstaller (DDU)\DDU v17.0.6.1\Display Driver Uninstaller.exe
    Faulting module path: C:\Windows\SYSTEM32\d3d9.dll
    Report Id: 4123ff90-ed4f-49c2-a973-ad99b7a5bea8
    Faulting package full name:

    Hope this helps Wagnard.

Your issue is related with the graphics drivers. D3d9.dll
Possibly the driver you currently have installed is not working correctly with the creator.

Then it could be an issue with the creative update. Another confirmed to me that other .Net applications, especially WPF one (like DDU) will have the same error.
WPF application uses the graphics card for acceleration.
Anyway, nothing I can do atm.

The real issue has been found (Thanks to a user on my forum).
Simply close MSI afterburner / RTSS or any application monitoring software like this one and all your .NET application that failed to launch will work now.

Fix for the above mentioned issue, all should work great now under Creators Update.

I was stuck during my install with this nvidia installer failed error and none of the fixes I had tried worked. I then came across this thread and figured, what the heck I have nothing to lose. I did a backup, ran the program and once it rebooted I tried to install my driver and BOOM! it worked first try!!! I just created this account to say THANK YOU, THANK YOU, THANK YOU! I was at my wits end and thought I was going to have to reload Windows. You guys saved me a huge PITA! :)

    hi!

I upgraded to the new win10 creators upgrade yesterday, as well as the new geforce driver 381.65.

I noticed some washed out look and less color in games and wanted to do a DDU uninstall/reinstall of the driver.

When I try to launch DDU in the new windows update I get the pop-up "let this program make changes to the pC" click yes, then a very quick blink and nothing happens, the program doesn't start.

Not sure if this is an issue with the windows update or just a local problem for me.

EDIT - read the above posts, closed MSI AB and it loaded fine!!!!!

Hmmm what does the Remove "GeForce Experience" display driver installer cache/files (Note: Drivers won't be removed) option do exactly? Should I use it even if I don't have GFE installed? Must be a new feature as I don't recall seeing it before (although I haven't run DDU in several months).

No need to run if you never installed GFE.
It only clean it's cache. By default, GFE doesn't remove the drivers installation files that it has downloaded.
